What is CAMEO™-96?

CAMEO™-96 is the first and only standardized colony-forming cell assay.

CAMEO™-96 combines a methylcellulose-based colony-forming cell (CFC) differentiation assay with the power of HALO® bioluminescence technology in a 96-well plate culture format. The same cultures that are used to manually enumerate colonies in the well are also used to measure the proliferation status of the cells. In short, both proliferation and differentiation are detected in the same culture, but using different readouts; proliferation is detected by measuring the intracellular ATP (iATP) concentration using bioluminescence technology, while differentiation is detected by counting the number of colonies in each well, prior to measuring bioluminescence. Since both the proliferation and differentiation assays are performed on exactly the same cultures using the same reagents and conditions, there is a direct correlation between the iATP concentration and the total number o colonies counted producing a positive linear regression. Because the iATP concentration measured is calibrated and standardized against an external ATP standard curve, the proliferation assay is used to standardize the CFC assay. As a result, the number of colonies counted can be read off the regression in iATP concentration equivalents. In addition, since the CFC assay correlates with a standardized ATP assay, it too has been standardized. As a result, CAMEO™-96 is the only standardized CFC assay available and provides the investigator with considerably more information that would otherwise be obtained with the CFC assay alone.
