












The CAMEO™ Platform
The CAMEO™ Platform from HemoGenix® consists of 2 different in vitro assays.
CAMEO™-4
CAMEO™-4 is a traditional colony-forming cell (CFC) assay that has been miniaturized so that 4 replicates of 0.1ml culture reagents are dispensed into a 4-well, 35mm Petri dish.
The total volume of the culture required for all 4 replicates is only 0.6ml. Unlike the traditional CFC assay for human cells, CAMEO™-4 using human target cells can be completed in just 7 days if required, rather than 14 days. This is because HemoGenix® determined in determined and showed in 2003 (Curr. Opin. Drug Disc. Develop. 6/1:100-109) that undifferentiated clusters or colonies of cells, designated "proliferation units" (PUs) counted at 7 days of incubation correlated not only with the proliferation of the cells, but also colonies of functionally mature cells counted at 10 or 14 days.

CAMEO™-96
CAMEO™-96 is the only in vitro assay that allows the user to detect and measure both proliferation and differentiation of lympho-hematopoietic stem and progenitor cells in the same assay.
For this reason, the assay has been given the name of CAMEO™-96, although proliferation occurs prior to differentiation. CAMEO™-96 is performed in a 96-well plate and uses exactly the same reagents as CAMEO™-4. The assay is performed in 2 stages. The first stage involves counting colonies in each well in a similar manner to the traditional CFC assay. After counting the colonies manually to assess differentiation, the proliferation status of the cells is determined using an ATP-based, luciferin/luciferase bioluminescence technology that allows the intracellular ATP (iATP) concentration to be measured in a plate luminometer. The iATP concentration is directly proportional to the proliferation status of the cells. Thus, both differentiation and proliferation are determined using the same cell culture, but different readouts.
