The HALO® Predictive Hemotoxicity Platform
HemoGenix® offers HALO® Predictive Hemotoxicity Platform in 3 formats.
HALO®-96 MeC (Methyl Cellulose) is similar to the traditional colony-forming assay in that it allows cells to grow under clonal conditions, but instead of manually counting colonies, an instrument-based, ATP bioluminescence readout is performed. The assay uses a 96-well plate format, but should really only be used for a small number of samples because the viscous methyl cellulose has to be dispensed manually. The HALO®-96 MeC Hemotoxicity Platform can be used for pre-clinical animal studies and patient monitoring during clinical trials.
HALO®-96 SEC (Suspension Expansion Culture) does not use methyl cellulose and therefore the whole assay, from beginning to end can be performed under fully automated conditions. The HALO®-96 SEC Hemotoxicity Platform is easier to use at any stage of drug discovery and development, from screening to patient monitoring during clinical trials. It is also 2-3 times more sensitive than the HALO®-96 MeC Hemotoxicity Platform.
HALO®-384 HT (High-Throughput) is similar to the HALO®-96 SEC Hemotoxicity Platform, but uses a 384-well plate format. It was specifically designed for high throughput screening at the ADME/Tox level and later and is even more sensitive than the HALO®-96 SEC Hemotoxicity Platform. HALO®-384 HT is the only high throughput hemotoxicity test system available.
For in-house hemotoxicity testing, HemoGenix® offers a large number of assay kit options.
The HALO®-DDI (Drug-Drug Interaction) Platform
HALO®-DDI is a new contract research service offered by HemoGenix®. It is not only the only hemotoxicity drug-drug interaction platform but the only drug-drug interaction platform that does not rely on the cytochrome P450 system, but rather invesigates the effects of two or more drugs at the cellular level. This provides significantly more predictive information at the organismic level than would otherwise be obtained.
HALO®-DDI is a 384-well plate based assay that utilizes HALO® SEC (Suspension Expansion Culture) technology. The usual cell population that is tested using HALO®-DDI is the mature multipotential stem cell population (CFC-GEMM), since this provide the most information on drug-drug interactions on the hematopoietic system. However, HALO®-DDI can also be performed on other lympho- hematopoietic cell populations upon request.
Part of the HALO®-DDI testing procedure is to perform a dose response on the individual compounds. The results may indicate little, if any, response to the lympho-hematopoietic system, a moderate response or a significant toxicity to the system. Even if the compound produces little, or no response, this does not mean that, when titrated against another compound, the two will not interact with each other. When interaction does occur, the usual response is that of a typical U-shaped dose response curve, especially at high doses.
HALO®-DDI is usually used to investigate the interaction of two compounds/drugs that may be administered to a patient in combination. However the interaction between more than two drugs can also be investigated.

Hands-On Training
HemoGenix® offers a comprehensive 3-4 day training course. During that time, you will learn the background of performing hemotoxicity testing using the HALO® Platform, you will get hands-on experience at setting up a complete experiment as well as how to process the plates and evaluate and interpret the results. You will have all the necessary experience to begin using the HALO® and our other ATP-based bioluminescence platforms.
