LUMENESC™ and Multiparameter Testing

LUMENESC

LUMENESC™, like the HALO® and LumiSTEM™ Platforms, utilizes a highly sensitive ATP-based bioluminescence signal detection system. Detection of intracellular ATP actually allows 4 different parameters to be detected or implied. These are:

  • Proliferation / Cytotoxicity
  • Viability
  • Mitochondrial integrity / Cellular Integrity
  • Apoptosis(see LUMENESC™ Applications, Mechanism of Action - Apoptosis).

In addition to these ATP-dependent parameters, a great deal more information can be obtained from the LUMENESC™ Platform, by simply increasing the number of replicate wells/sample. By increasing the number of wells initially prepared, multiple parameters can be obtained from the same sample. For instance, if instead of the normal number of wells prepared to detect proliferation or cytotoxicity, double the number of wells is set up, then half can be used for the proliferation/cytotoxicity assay, while the other half can be removed and the cells used for phenotyping. The supernatant from the individual or pooled cultures can be analyzed for growth factors, cytokines or other soluble factors using ELISAs etc. If more replicate wells are prepared, then the remainder can be used for gene expression analysis. In this way, a large amount of information can be obtained from a single sample.

The LUMENESC™ Platform is also part of the ComparaTOX™, Predictive In Vitro, Cross-Platform, Comparative Toxicity Testing System developed by HemoGenix®. ComparaTOX™ combines the HALO® Predictive Hemotoxicity Platform and the LumiSTEM™ Platform for other proliferating cell types, with LUMENESC™ to provide a unique and extremely powerful comparative, ranking toxicity testing system for multiple cell types.
