











The LumiSTEM™ Platform
for Primary Stem and Progenitor Cells and Cell Lines


LumiSTEM™ is the third assay system developed by HemoGenix® that uses the highly sensitive, non-subjective and fully standardized ATP-based luminescence signal detection system originally developed for the HALO® Platform.
LumiSTEM™ allows investigators to measure proliferation (and in the case of LumiCLONE, differentiation) of many different cell types using their own culture reagents and protocols. The types of cells that can be used with LumiSTEM™ include:

LumiSTEM™ Assays for Specific Applications
LumiSTEM™-96 is a proliferation / cytotoxicity assay performed in a 96-well plate. It can be used to study the effect of virtually all agents on the cell types listed above. Such agents might include drugs, chemicals, radiation, growth factors and cytokines to mention just a few. LumiSTEM™-96 complements and can be used in conjunction with all HALO®-96 platforms for lympho-hematopoietic cells and LUMENESC™-96 for mesenchymal stem cells.
LumiSTEM™-96 PREP is a secondary re-plating assay designed to detect and substantiate stem cell self-renewal and measure stem cell expansion potential. LumiSTEM™-96 PREP complements HALO®-96 PREP for lympho-hematopoietic cells.
LumiCLONE-96 allows the investigator to grow cells under clonogenic conditions in methylcellulose. After the incubation period has elapsed, the colonies are counted in each well. This provides a visual and manual method of assessing differentiation of the target cells. The contents of each well is then processed using a luciferin/luciferase reagent to measure intracellular ATP, the concentration of which is directly correlated to the proliferation status of the cells. The assay therefore allows both cell proliferation and differentiation to be measured using the same sample, but with two different readouts. An additional advantage of this assay is that counting the colonies and plotting the number of colonies against the ATP concentration should produce a correlation. This correlation allows colony number to be expressed as standardized ATP concentrations. LumiCLONE-96 complements and can be used in conjunction with CAMEO™-96 DP for lympho-hematopoietic cells.
LumiSTEM™-384 HT is a high-throughput proliferation/cytotoxicity screening platforms for drug candidates and environmental agents. This assay requires the use of a liquid handler. LumiSTEM™-384 HT compliments and can be used in conjunction with HALO®-384 HT for lympho-hematopoietic cells and LUMENESC™-384 HT for mesenchymal stem cells.
LUMINOMETERS
A plate luminometer is not the same as that for measuring absorbence or fluorescence. Multi-parameter instruments are available that can measure, absorbence, fluorescence and luminescence. Alternatively, a dedicated luminometer that only detects luminescence (both chemi- and bioluminescence) can also be obtained.
