What is Cell Potency?
  • Cell potency is the quantitative measurement of cell proliferation potential. This, in turn, is a measure of cell primitiveness and cell engraftment potential.

(1): If the MSC source is not cord blood or bone marrow, use MSCGlo™-RS to establish a reference standard (RS) for that tissue source. A RS must always accompany a sample when potency is measure, since the potency ratio can only be determined by comparsion of the sample with a RS.

(2): One RS must always be performed when an unknown sample is measured. If more than 1 sample is to be measured, a single RS of the same cell type can be used to compare with all samples performed on the same day.

Difference between MSCGlo™-Potency and MSCGlo™-RS

To measure potency, a reference standard of the same material as the sample is required so that the potency ratio can be determined. The inclusion of a reference standard to measure potency is an FDA requirement. Universal or global reference standards for cell therapy products are not available. As a result, it is necessary for individual centers to establish ther own reference standards. To accomplish this, a standardized and validated assay platform is required.

MSCGlo™-RS is used to establish your first in-house reference standard. The assay includes a vial of cord blood, peripheral blood or bone marrow cryopreserved cells that are used to compare to your own preparation. MSCGlo™-RS is only required once to set up your first reference standard cell preparation. All further reference standards establish in-house will be compared to the previous reference standard. (See below).

To measure potency of your samples prior to use, MSCGlo™-Potency is used. MSCGlo™-Potency does not include reference standard material. You will use an in-house reference standard to measure the potency ratio of the sample.

MSCGlo™-Real Time is the first assay to measure MSC proliferation in real time and to follow MSC growth kinetics in culture.

MSCGro™-Real Time uses a non-destructive bioluminescence luciferase marker that is reduced in the mitochondria of cells. As the cells proliferate, the biochemical reduction process increases proportionately with an increase in bioluminescence.

 How MSCGlo™-Real Time is Used

The MSCGlo™-Real Time Assay is simple. Obtain MSCs from any source and adjust the cell concentration with MSCGro™ (provided). Mix in the MSCGlo™-Real Time reagent and dispense the Culture Mast Mix into the wells of the 96-well plates provided with the kit. Measure the bioluminescence at time 0 in a luminescence plate reader and transfer the plate to a 37oC humidified incubator gassed with 5% CO2 and, if possible, 5% O2. (MSC demonstrate greater plating efficiency at 5%O2, than in atmospheric conditions). The plate can be removed from the incubator at any time to measure the luminescence. Depending on the initial number of cells plated/well, MSC growth should maximize over a 2-3 day culture period. 

At any time, fluorochrome-conjugated antibodies to MSCs or other cells (for example CD73, CD90, CD105, CD106, CD34) can be added. If the flow cytometer is equipped to measure cells in a 96-well plate, the phenotype of the cells can be determined directly. If this option is not available, then the cells can be removed from one or more of the replicate wells and the antibodies added after transferring the cells to a tube for phenotypic analysis.

The MSCGlo™-Real Time Reagent need not be added at the beginning of the culture. MSCGlo™-Real Time can also be used as an endpoint assay, i.e. to measure the amount of MSC prodiferation at the end culture. This would be similar to MSCGlo™-96 Research. In contrast to MSCGlo™-Real Time, however, MSCGlo™-96 Research is a a "destructive" (cells are lysed) ATP bioluminescence assay that is fully standardized assay and includes controls and standards in the assay kit.The results between MSCGlo™-Real Time and MSCGlo™-96 Research correlate with each other since both measure MSC proliferation, but in different ways. MSCGlo™-96 Research is usually more senitive than MSCGlo™-Real Time.
