The HALO® 7-Population Predictive Hemotoxicity Platform
The HALO® 7-Population Predictive Hemotoxicity Platform is a unique assay designed to examine the response of 7 lympho-hematopoietic cell populations simultaneously to one or more compounds or agents tested on up to 5 different species (human, non-human primate, dog, rat and mouse).
The 7 populations are:
2 Stem Cell Populations
- HPP-SP, a High Proliferative Potential - Stem and Progenitor cell population that is primitive enough to produce both lympho- and hematopoietic cells, and
- CFC-GEMM (Colony-Forming Cell - Granulocyte, Erythroid, Macrophage, Megakaryocyte), which is a mature stem cell population capable of producing cells of the 3 hematopoietic lineages, the erythroid lineage, the myelomonocytic lineage (consisting of both the granulocyte and macrophage lineages) and the megakaryocytic lineage.
3 Hematopoietic Cell Populations
- BFU-E, Burst-Forming Unit - Erythroid, the primitive erythropoeitic progenitor cell,
- GM-CFC, Granulocyte-Macrophage Colony-Forming Cell, the primitive myelomonocytic progenitor cell and,
- Mk-CFC, Megakaryocyte Colony-Forming Cells, the primitive megakarocytic progenitor cell.
2 Lymphopoietic Cell Populations
- T-CFC, the T-lymphocytic progenitor cell and,
- B-CFC, the B-lymphocytic progenitor cell.
The analysis of the results consists of 3 stages:
By analyzing the response of all 7 populations simultaneously, a "global" view of the the differential effects of one or more compounds detected on one or multiple species can be ascertained and ranked in order of toxicity.
However, this is just the first stage in the analysis. The HemoGenix® Predictive Hemotoxicity Paradigm is then employed, in the second stage, to predict the effects to the whole lympho-hematopoietic system.
The third stage in the analysis is to predict the estimated LD50 followed by the in vivo doses in both mg/kg and mg/m2 for the IC50, IC75 and IC90 values. The IC90 values provide the estimated Maximum Tolerated Dose (MTD).
Which HALO® 7-Population Predictive Hemotoxicity Platform should be used?
The HALO® 7-Population Predictive Hemotoxicity Platform is available as a contract research service or as assay kits using the HALO®-96 MeC, HALO®-96 SEC or HALO®-384 HT format.
If the HALO® 7-Population Predictive Hemotoxicity Platform is to be used for a small number of lead compounds, we suggest using either HALO®-96 MeC or HALO®-96 SEC. If, on the other hand, large numbers of compounds have to be tested on multiple species, we suggest either the HALO®-96 SEC or HALO®-384 HT format.
