











LumiSTEM™ is the third proliferation assay system developed by HemoGenix® that uses the highly sensitive, non-subjective and fully standardized ATP-based luminescence signal detection system originally developed from the HALO® Platform.
LumiSTEM™ is for investigators who want to measure the proliferative / cytotoxic status of non-hematopoietic stem cells and other proliferating cell types under varying conditions.
How LumiSTEM™ Works
Cells can be grown using the investigator’s normal protocols and transferred to the 96- or 384-well plate(s) provided. Alternatively, cells can be grown directly as non-adherent or as adherent cells in the 96- or 384-well plates provided with the kit. In addition, cells can be grown under clonal conditions in 96-well plates using the semi-solid methyl cellulose supplied in each kit. Growth factors are not included and must be provided by the investigator. The cells are cultured according to the investigator’s protocols. During proliferation, the cells increase their intracellular ATP concentrations. At the end of culture, the intracellular ATP is released and acts as a limiting substrate for a luciferin / luciferase reaction that produces bioluminescence in the form of light which is measured in a plate luminometer (supplied by investigator). Standardization of LumiSTEM™ is accomplished by performing an ATP dose response. LumiSTEM™ is easy-to-use, rapid, reproducible and reliable.
Characteristics of the new, 3rd Generation Luminescence Reagent
LUMINOMETERS
A plate luminometer is not the same as that for measuring absorbence or fluorescence. Multi-parameter instruments are available that can measure, absorbence, fluorescence and luminescence. Alternatively, a dedicated luminometer that only detects luminescence (both chemi- and bioluminescence) can also be obtained.
